Is there any difference in power between as assault blaster used by the clones and someone like Padme's blaster which is just a pistol.

I only say because both seem to kill droids in one hit and from multiple ranges.

not the size of the blaster that counts

it's what you do with it

    apparently :p

Or who shoots it (plot necessity).

I think there are differences when it comes to bigger targets, who have better armor. Normal droids (B1) can be killed with one shot.
However I think that it takes more shots to bring down a B2 droid with just the pistol.

I think that assault rifles deal more damage in case of well armored targets.
The scene below comes to mind (from ROTS):

I don't think that you can kill a large crab-droid with a blaster pistol.

Assault rifles also have a greater fire rate, which is another advantage.

Blaster rifles and heavy blaster pistols pack more punch than a standard blaster pistol.

Well when I play KOTOR the pistols are a 1-6 for damage and the blaster rifles are a 1-8 :)

The movies (and TCW) seem to indicate that larger blasters fire larger bolts that seem to have more "force" behind them, in addition to being able to fire more bolts in succession (like the assault rifles with a greater fire rate mentioned above).

I'd assume it works in a similar manner as bullets, rifles have better range and power and higher ammunition capacity.

Of course there's going to be outliers. Han's blaster seemed to be powerful enough to take chunks out of granite pillars and hit stormtroopers with shrapnel. So I'd say it's somewhat equivalent of a magnum or 50 caliber pistol.

Granite? Docking Bay 94 was described as a dirt pit in the novel.

The DL-44 (Han's blaster) is a heavy blaster pistol so they are pretty powerful.
