Are you talking about Aayla Secura? She was in Attack of the Clones, and I'm almost positive she's supposed to be in Revenge of the Sith as well.

 

If you're talking about the red "Twi'Lek", she's not a twi'lek. She's Shaak Ti, a Togruta also seen in AOTC.


You're not the first person to mistake her for a twi'lek.

No, during the ROTS council seat several chairs are empty.
She is not present during any of the scenes.

 

Shaak Ti is in the council scenes, though her part was shot seperately. There is only one seat completely empty (some are filled with holograms), and that doesn't stay empty for very long.
