In ANH, in the final (I think) draft of the script, Vader is supposed to say: 'And bring me the ambassador, I want her alive!', but in the movie it sounds like 'bring me the passengers, I want them alive.' Which is it?

And if he did say 'ambassador', how did he know it was a she?

I'm pretty sure he does say "passengers" in the film.

As for "ambassador," he's already familiar with Princess Leia traveling on board the Tantive IV, so he would rightfully know the ambassador is a she.

Definitely NOT ambassador! He clearly says "bring me the passengers, I want them alive!"
Also remember that Vader didn´t believe Cpt Antilles when he said that it was a consular(sp?) ship....

I just checked it out. First he says, "If this is a counsilor ship, where is the ambassador?" then he says, "...bring my the passengers, I want them alive." Closed-captioning confirms it.

I read the first post wrong. He says he wants the passengers. I really need to pay attention when I'm reading.

    You're all wrong.
He says "And bring me a basset hound."

It happens that the ambassador is a passenger, so it works regardless. :p
