    Originally, as of the end of Season 2 - beginning of Season 3, to make Nala Se cooperate, because she wouldn't after the destruction of Kamino. In exchange for his freedom, Lama Su told Hemlock that Nala Se has developed an attachment to Omega, and that would make her cooperate.
    In Season 1, Lama Su wanted to bring Omega back, because she is a pure genetic replica of Jango Fett, which means they could create new and superior clones, since the genetic material from Jango had long deteriorated to create anything good. Kaminoans were hoping to negotiate with the Empire so that they wouldn't terminate the contracts by creating a new, superior clone. He told Nala Se to bring Omega back, get the genetic material needed from her, and then terminate her. Lama Su hired Cad Bane to retrieve Omega. Nala Se hired Fennec Shand in secret to mess with Lama Su's plan, since she didn't want Omega back and terminated.

    Only Nala Se originally knew why Omega was important and how she was different, since she created her. But she had developed an attachment to her, and since then she kept protecting her, which is why she let her escape with Clone Force 99 in the very first episode, protected her by hiring Fennec Shand, and now she kept destroying her blood samples so that whatever it is that makes her special in her DNA would not be exposed to the Empire — since that would mean Omega's certain death. But now Emerie tested Omega's blood and reported the successful midi-chlorian transfer without degradation, which is the key to Hemlock's research. So now Hemlock with the full resources of the Empire is after her in order to complete the Emperor's project.

    So basically Palpatine wants clones of himself, but the clones don't have enough midichlorians? I guess this is where Rey's father comes from?

    So then what do they do, mix Palpatine's blood with Omega's, and that does the trick? They don't need to kill Omega for that, do they? In fact, wouldn't they want to keep her alive?

    Since we see Palpatine does create a Force-sensitive clone of himself in TROS, I guess that means they succeed in recapturing Omega?

    Why does Palpatine need Force-sensitive clones of himself, why not just find some Force-sensitive individual and transfer his essence/soul into them? And why is he interested in gaining Luke as an apprentice if his cloning program succeeded?

    I haven't seen TLJ and TROS and haven't read any TROS-related material, so I dunno what they did there, beyond "somehow Palpatine returning", so I will rely only on The Bad Batch when discussing The Bad Batch. From what I understood, Palpatine either wants to create clones that have as much midi-chlorians as the source, or transfer midi-chlorians into someone (the clones probably). The problem is that midi-chlorian count is severely reduced in the process, and when transferring they degrade, so you get an extremely weak being when it comes to the Force potential.
    I doubt it was Palpatine's blood. They have "specimens" locked up in the vault at Tantiss, that Sith-temple-looking room, where Palpatine and Hemlock went. Could be dead/in stasis Jedi that are harvested for midi-chlorians — but that's just a guess, I am sure they will reveal it all later. But blood is just for initial testing to see if its viable at all. Ultimately, they want to find out how to make midi-chlorian transfer successful, which means they will have to cut Omega open and study her genetic material by harvesting all of it. That study will have to go beyond simple blood sample, I must imagine. They don't need Omega alive. They just need to find out the key in her DNA so that they can replicate it in their own research.
    If I recall correctly, they still work on Project Necromancer in The Mandalorian S3. So, Palpatine didn't return yet by that time. We also know that Gideon was hunting down Grogu for the same purpose that Hemlock wants Omega, but Gideon wanted it to create Force sensitive clones of himself. Which means he knew how to achieve that by having Grogu for his blood, which means the knowledge had been obtained before him. So they will get something out of Omega by the end of the season. We'll have to wait and see. I doubt Omega will die, that would be way too grim for Star Wars.
    Well, it didn't fully succeed yet as of ROTJ, apparently. As for why not transfer his essence/soul into them, I dunno, maybe there are some complications with that, and transferring it into a clone, which is just an empty vessel, is easier.

    This is just an aside but why does Emerie Karr have a last name? She is one of three clones to have a last name.

    also where did she get her doctorate? Was it from an accredited source or was Hemlock like hey you have followed me around enough your a doctor now.

    First, I think he means clones of Jango. And second, that's a Legends character. Not canon, like specified.

    No official explanation yet, but it seems like there are some possibilities:
    1) Like Cut, she is married.
    If that is the case, I wouldn't be surprised if Hemlock is holding her spouse hostage for her good behavior, like how he used Omega against Nala Se and Crosshair against Omega.
    2) They wanted to disguise that she is a clone for some reason. After all, she isn't one of the troopers but she was given the growth exhilaration, so the reason for her creation is certainly mysterious.
    3) Just an affection, like a clone nickname with a bit more added on.

    Looking at her face, she does share a lot of features with the other Jango clones, particularly when she doesn't have the glasses on:

    So i don't think she's Emeerie Karr, but rather that Hemlock fabricated her whole identity to hide the fact that she's a clone, including her name and surname.

    Also she’s straight up played by a Māori women. Well half anyway.

    which fun fact did you know the actress who voices Emerie Karr also was the queen of Naboo for the funeral scenes in ROTS

    it’s why I didn’t get why anyone didn’t completely understand she was a clone the moment we heard her voice. Almost every single actor or actress from New Zealand in Star Wars has either played a clone or a clone adjacent role. There are some exceptions, but for actual speaking roles almost every single actress or actor who has appeared in Star Wars and is from New Zealand has been tangentially related to cloning. Her being native just confirmed in my head that she was a clone. But then again people don’t pay attention to credits as much as me. :p

    by the way go watch the film Whale Rider which stars her and she was nominated for best actress over. Really good film.

    I didin't know about all of those casting details before, nice catch, this just makes it more apparent that she is a Jango clone, i wonder if we will get more exploration of her backstory and in what way Hemlock influenced her.
